  • Heat dry Magnesium Sulfate on low heat.
  • Bubble Methylamine gas through Methanol.
  • Add heated Magnesium Sulfate to the Methanol solution and stir for 5 minutes.
  • Filter the anhydrous Methanol / Methylamine solution from the Magnesium Sulfate precipitate.
  • Add 300g of P2P to an anhydrous solution with strong magnetic stirring.
  • Leave the mixture to mix for 1 hour.
  • You will observe a cloudy color change during this time due to Imine formation.
  • Pipet out the lower clear layer and discard it.
  • Cool the solution using dry ice or an ice bath and magnetic mixing.
  • Add 10mg of Sodium Borohydride while maintaining cooling. It is crucial to prevent the temperature from rising and the flask should not feel warm.
  • Repeat the process and add another 10mg until you have added a total of 35 mg to cool it further. Be patient and take your time with this step, as it is prone to failure.
  • Allow the solution to stir for 1 hour.
  • Mix thoroughly 1 liter of DCM with 7 liters of distilled water.
  • Add P2P solution and mix for 5 minutes.
  • Let the DCM settle at the bottom, leaving water on top.
  • Pour off the water carefully, leaving only the DCM oil. You might lose some minimal quantity of the product while drawing off the water.
  • Bubble HCl gas into DCM solution until pH is less than 6.0.
  • Evaporate solution on hotplate until dry.

  • Stir and cool to 0°C.
  • Gradually add NaBH4 and stir for 1 hour.
  • Increase temperature to 40°C for 30 minutes.
  • Remove 2/3 of MeOH through distillation.
  • Add KOH and extract with toluene.
  • Treat with HCl gas.

 

Is MeOH really that much more efficient?

 

Yes, you can run a slurry reaction with DCM. However, it is more efficient and produces higher yields to run it in MeOH. DCM reacts with the amine to form the n-chloro ammonium salt, which is undesirable. By drying with MgSO4, you can even use aqueous methylamine instead of methanoic.
